Amisulpride Tablets I.P.

Varied (Amisulpride Tablets I.P.) contains the antipsychotic salt amisulpride, a selective dopamine D₂/D₃ receptor antagonist with relative preference for pathways in the limbic system. By dampening overactive dopamine signaling, it reduces hallucinations, delusions, agitation and thought disorganization. At lower exposures, amisulpride also blocks presynaptic autoreceptors, which can modestly enhance dopamine release in underactive circuits and help with “negative” symptoms such as apathy, social withdrawal and blunted affect. Compared with older antipsychotics, amisulpride has minimal anticholinergic effects, but like others in its class it can cause movement symptoms and often raises prolactin.

Varied (amisulpride) is prescribed for schizophrenia across acute episodes and maintenance. Clinicians may also use lower doses to target negative symptoms or persistent mood-related features when amisulpride is considered appropriate.

The three strengths 50 mg, 100 mg and 200 mg allow precise titration. Treatment is individualized: doctors usually start low and adjust in small steps until symptoms are controlled with good tolerability. Tablets are taken once or twice daily at the same time each day with water. Do not stop suddenly unless your doctor advises a taper, and do not change the dose on your own. Older adults and people with kidney problems generally start at lower doses with slower adjustments. If a dose is missed, take it when remembered unless it is close to the next dose; do not double up.

Storage: Store Varied in a cool, dry place away from moisture and sunlight, and keep out of the reach of children.

Common Side Effects: Some patients notice sleepiness, dizziness, restlessness or akathisia, tremor, muscle stiffness, dry mouth, constipation, nausea or insomnia. Weight gain and increases in blood sugar or lipids can occur. Because amisulpride commonly elevates prolactin, women may experience breast tenderness, missed periods or milk leakage, and men may notice decreased libido or erectile difficulty; report these to your clinician. Seek urgent help for symptoms suggesting neuroleptic malignant syndrome (high fever, severe muscle rigidity, confusion), severe allergic reaction, a very fast or irregular heartbeat, fainting, or uncontrollable movements of the face or tongue.

Drug Warnings: Before taking Varied (amisulpride), inform your doctor if you have high or low blood pressure, heart-rhythm problems or a family history of long QT, low potassium or magnesium, thyroid disease, diabetes or high cholesterol, Parkinson’s disease, seizure disorders, kidney impairment, prolactin-dependent tumors (such as a prolactinoma or breast cancer history), or depression with suicidal thoughts. Avoid taking this medicine with other sedating medications unless advised, as it may increase drowsiness and impair alertness.

Drug–Drug Interactions: Do not combine amisulpride with levodopa or dopamine agonists because it counteracts their effect. Use caution with medicines that prolong the QT interval (certain antiarrhythmics, macrolide and fluoroquinolone antibiotics, some antihistamines and antipsychotics) or with drugs that lower potassium or magnesium (loop/thiazide diuretics, high-dose laxatives), as these raise the risk of serious arrhythmias. Central nervous system depressants alcohol, benzodiazepines, opioids and sleep medicines can add sedation. Because amisulpride is cleared mainly by the kidneys, medicines that markedly affect renal function may influence exposure; your doctor will monitor as needed. Always share a complete list of prescription and over-the-counter drugs and supplements.

Drug–Food Interactions: Varied can be taken with or without food. Alcohol should be limited or avoided, as it increases drowsiness and slows reaction time. There are no specific dietary restrictions.

Drug–Disease Interactions: Patients with kidney impairment require lower doses and careful monitoring. Caution is advised in people with cardiovascular disease, electrolyte abnormalities, diabetes/metabolic syndrome, Parkinson’s disease, seizures, and those at risk for venous thromboembolism or stroke. Use in elderly patients with dementia-related psychosis is associated with increased mortality and is not approved in many regions.
